This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. However, to date, only gross deletions of the CATSPER2 gene have been reported in association with human disease. The R498X variant is observed in 19/277,114 total alleles in large population cohorts (Lek et al., 2016). We interpret R498X as a variant of uncertain significance. -
