The synthesis of thyroid hormone is catalyzed by a protein complex located at the apical membrane of thyroid follicular cells. This complex contains an iodide transporter, thyroperoxidase, and a peroxide generating system that includes this encoded protein and DUOX1. This protein is known as dual oxidase because it has both a peroxidase homology domain and a gp91phox domain. [provided by RefSeq, Jul 2008]

This variant is present in population databases (rs119472029, gnomAD 0.02%). This missense change has been observed in individual(s) with congenital hypothyroidism (PMID: 16134168, 22336364, 30240412). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 4065). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt DUOX2 protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects DUOX2 function (PMID: 17374849). For these reasons, this variant has been classified as Pathogenic. - |
