This enzyme is involved in creatine biosynthesis, whereby it catalyzes the transfer of a guanido group from L-arginine to glycine, resulting in guanidinoacetic acid, the immediate precursor of creatine. This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 23 of the GATM protein (p.Arg23Pro). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GATM-related conditions.
