Genes affected
TPM1 (HGNC:12010): (tropomyosin 1) This gene is a member of the tropomyosin family of highly conserved, widely distributed actin-binding proteins involved in the contractile system of striated and smooth muscles and the cytoskeleton of non-muscle cells. Tropomyosin is composed of two alpha-helical chains arranged as a coiled-coil. It is polymerized end to end along the two grooves of actin filaments and provides stability to the filaments. The encoded protein is one type of alpha helical chain that forms the predominant tropomyosin of striated muscle, where it also functions in association with the troponin complex to regulate the calcium-dependent interaction of actin and myosin during muscle contraction. In smooth muscle and non-muscle cells, alternatively spliced transcript variants encoding a range of isoforms have been described. Mutations in this gene are associated with type 3 familial hypertrophic cardiomyopathy and dilated cardiomyopathy 1Y. [provided by RefSeq, Jun 2022]

Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Sep 06, 2017 | A variant of uncertain significance has been identified in an alternate transcript of the TPM1 gene. The R191X variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). R191X is predicted to cause loss of normal protein function either by protein truncation or nonsense-mediated mRNA decay. However, loss of function is not a known mechanism of disease for the TPM1 gene. Furthermore, no nonsense variants in this transcript of the TPM1 gene have been reported in Human Gene Mutation Database (Stenson et al., 2014). - |

TPM1-related disorder Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | PreventionGenetics, part of Exact Sciences | Oct 02, 2023 | The TPM1 c.571C>T variant is predicted to result in premature protein termination (p.Arg191*). To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0062% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/15-63353404-C-T). Of note, loss of function variants have not commonly been reported in the TPM1 gene. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. - |
