Genes affected
MTFMT (HGNC:29666): (mitochondrial methionyl-tRNA formyltransferase) The protein encoded by this nuclear gene localizes to the mitochondrion, where it catalyzes the formylation of methionyl-tRNA. [provided by RefSeq, Jun 2011]

Pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpJan 11, 2024This sequence change creates a premature translational stop signal (p.Arg128*) in the MTFMT gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MTFMT are known to be pathogenic (PMID: 21907147, 24461907). This variant is present in population databases (rs397514613, gnomAD 0.002%). This premature translational stop signal has been observed in individual(s) with combined oxidative phosphorylation deficiency (PMID: 21907147). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 39828). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
