Also referred to as Batten disease, NCL comprises a class of autosomal recessive, neurodegenerative disorders affecting children. The genes responsible likely encode proteins involved in the degradation of post-translationally modified proteins in lysosomes. This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 201 of the CLN6 protein (p.Lys201Glu). This variant is present in population databases (rs587780316, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with CLN6-related conditions. ClinVar contains an entry for this variant (Variation ID: 128787). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
