Also referred to as Batten disease, NCL comprises a class of autosomal recessive, neurodegenerative disorders affecting children. The genes responsible likely encode proteins involved in the degradation of post-translationally modified proteins in lysosomes. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a CLN6-related disease. While this variant is not present in population databases (ExAC), the frequency information is unreliable due to low sequence coverage at this site. In summary, this is a novel in-frame insertion with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. -
