An increased activity of the encoded protein can result in an increased folate turnover rate and folate depletion. This variant is present in population databases (rs138155613, gnomAD 0.1%), and has an allele count higher than expected for a pathogenic variant. This sequence change affects codon 126 of the MTHFS mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the MTHFS protein. It affects a nucleotide within the consensus splice site. -
