WDR73 (HGNC:25928): (WD repeat domain 73) The protein encoded by this gene is thought to contain multiple WD40 repeats. WD40 repeats are motifs that contain 40-60 amino acids, and usually end with Trp-Asp (WD). This protein is found in the cytoplasm during interphase, but accumulates at the spindle poles and astral microtubules during mitosis. Reduced expression of this gene results in abnormalities in the size and morphology of the nucleus. Mutations in this gene have been associated with Galloway-Mowat syndrome PMID: 25466283), which is a rare autosomal recessive disorder that affects both the central nervous system and kidneys. The observed missense c.1126G>A (p.Ala376Thr) variant in WDR73 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Ala376Thr variant is absent in gnomAD Exomes. This variant has been submitted to the ClinVar database as Uncertain Significance. The amino acid change p.Ala376Thr in WDR73 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Ala at position 376 is changed to a Thr changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as a Variant of Uncertain Significance (VUS). In absence of another reportable variant in WDR73 gene, the molecular diagnosis is not confirmed. -

This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 376 of the WDR73 protein (p.Ala376Thr). This variant is present in population databases (no rsID available, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with WDR73-related conditions. ClinVar contains an entry for this variant (Variation ID: 1951533).
