The c.2131A>T (p.N711Y) alteration is located in exon 21 (coding exon 20) of the FANCI gene. This alteration results from a A to T substitution at nucleotide position 2131, causing the asparagine (N) at amino acid position 711 to be replaced by a tyrosine (Y). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Fanconi anemia Uncertain:1
This sequence change replaces asparagine with tyrosine at codon 711 of the FANCI protein (p.Asn711Tyr). The asparagine residue is weakly conserved and there is a large physicochemical difference between asparagine and tyrosine. This variant is present in population databases (rs766073258, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with FANCI-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
