This alteration results from a A to G substitution at nucleotide position 1403, causing the asparagine (N) at amino acid position 468 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

The POLG c.1403A>G variant is predicted to result in the amino acid substitution p.Asn468Ser. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0046% of alleles in individuals of European (non-Finnish) descent in gnomAD. An alternative nucleotide change affecting the same amino acid (p.Asn468Asp) has been reported in individuals with autosomal recessive POLG-related mitochondrial disease (Luoma et al. 2004. PubMed ID: 15351195). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
