This sequence change replaces leucine, which is neutral and non-polar, with histidine, which is basic and polar, at codon 312 of the NTHL1 protein (p.Leu312His). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with NTHL1-related conditions. ClinVar contains an entry for this variant (Variation ID: 823190). The p.L312H variant (also known as c.935T>A), located in coding exon 6 of the NTHL1 gene, results from a T to A substitution at nucleotide position 935. The leucine at codon 312 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
