Tuberin interacts with hamartin to form the TSC protein complex which functions in the control of cell growth. This TSC protein complex negatively regulates mammalian target of rapamycin complex 1 (mTORC1) signaling which is a major regulator of anabolic cell growth. Adequate data is not available in large population cohorts to assess the frequency of this variant in publicly available databases. Several in-silico splice prediction models predict that c.-30+5 G>T may destroy the natural donor site for exon 1 and lead to abnormal gene splicing. However, exon 1 is noncoding and in the absence of RNA/functional studies, the actual effect of this sequence change in this individual is unknown. Therefore, based on the currently available information, it is unclear whether this variant is a pathogenic variant or a rare benign variant. -
