Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TSC2 protein function with a negative predictive value of 95%. The p.S664C variant (also known as c.1991C>G), located in coding exon 18 of the TSC2 gene, results from a C to G substitution at nucleotide position 1991. The serine at codon 664 is replaced by cysteine, an amino acid with dissimilar properties. This variant was detected as homozygous in at least one individual (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
