The valine at codon 705 is replaced by methionine, an amino acid with highly similar properties. An in vitro functional assay showed that this variant is deleterious compared to wild-type (Hoogeveen-Westerveld M et al. Hum Mutat 2013 Jan;34(1):167-75). This alteration has been observed in at least one individual with a personal and/or family history that is consistent with TSC2-related disease (Personal communication).
