In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with TSC2-related disease. This sequence change replaces threonine with methionine at codon 1023 of the TSC2 protein (p.Thr1023Met). The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and methionine. This variant is not present in population databases (ExAC no frequency). -

The p.T1023M variant (also known as c.3068C>T), located in coding exon 26 of the TSC2 gene, results from a C to T substitution at nucleotide position 3068. The threonine at codon 1023 is replaced by methionine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
