Variant summary
Our verdict is Likely benign. The variant received -4 ACMG points: 2P and 6B. PM1BP4BP6BS2
The NM_000548.5(TSC2):āc.3199G>Cā(p.Val1067Leu) variant causes a missense change. The variant allele was found at a frequency of 0.00000372 in 1,613,030 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Synonymous variant affecting the same amino acid position (i.e. V1067V) has been classified as Likely benign.

The p.V1067L variant (also known as c.3199G>C), located in coding exon 27 of the TSC2 gene, results from a G to C substitution at nucleotide position 3199. The valine at codon 1067 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
