Tuberous sclerosis 2 Uncertain:3Benign:1
The TSC2 c.5297C>T p.(Pro1766Leu) missense change has a maximum subpopulation frequency of 0.0029% in gnomAD v2.1.1 (https://gnomad.broadinstitute.org). The in silico tool REVEL is inconclusive about a pathogenic or benign effect of this variant on protein function, and to our knowledge functional studies have not been performed. To our knowledge, this variant has not been reported in individuals with tuberous sclerosis complex. In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has therefore been classified as of uncertain significance.
This TSC2 missense variant (rs748936673) is rare (<0.1%) in large population dataset (gnomAD v.4.1.0: 6/1612950 total alleles, 0.0004%, 0 homozygotes), and has been reported in ClinVar (Variation ID: 535888). Two bioinformatic tools queried predict that this variant would be likely damaging to the protein, and the proline residue at this position is evolutionary conserved in most species assessed. We consider the clinical significance of c.5297C>T in TSC2 to be uncertain at this time.
Tuberous sclerosis syndrome Uncertain:1
This missense variant replaces proline with leucine at codon 1766 of the TSC2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with TSC2-related disorders in the literature. This variant has been identified in 1/250264 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
Lymphangiomyomatosis;C1846385:Isolated focal cortical dysplasia type II;C1860707:Tuberous sclerosis 2 Uncertain:1
not provided Uncertain:1
In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; Has not been previously published as pathogenic or benign to our knowledge
Hereditary cancer-predisposing syndrome Uncertain:1
The p.P1766L variant (also known as c.5297C>T), located in coding exon 41 of the TSC2 gene, results from a C to T substitution at nucleotide position 5297. The proline at codon 1766 is replaced by leucine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
