Though this exact alteration has not been reported in the literature, two other alterations at this same position (c.2834+1G>A and c.2834+1G>T) have been reported in probands with breast cancer (Susswein LR et al. Genet. Med., 2016 08;18:823-32; Antoniou AC et al. N. Engl. J. Med. 2014 Aug;371(6):497-506). This nucleotide position is highly conserved in available vertebrate species. Using the BDGP and ESEfinder splice site prediction tools, this alteration is predicted to weaken the efficiency of the native splice donor site; however, direct evidence is unavailable. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as likely pathogenic. -
