This changes the amino acid from a tryptophan to a stop codon within coding exon 7. This alteration was detected in 1/5589 German BRCA1/2-negative probands with breast cancer (Hauke J et al. Cancer Med, 2018 04;7:1349-1358). This alteration was also detected in a patient with a malignant peripheral nerve sheath tumor. The patient also carried a gross deletion in the NF1 gene (Byrjalsen A et al. PLoS Genet, 2020 12;16:e1009231). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
