Following imputation in a set of 166K Icelanders (239 imputed heterozygotes) we observed an association with breast cancer using 6908 cases and 292623 controls (OR= 3.58, P= 1.46e-06). The p.W906* pathogenic mutation (also known as c.2718G>A), located in coding exon 7 of the PALB2 gene, results from a G to A substitution at nucleotide position 2718. This changes the amino acid from a tryptophan to a stop codon within coding exon 7. This alteration has been reported in multiple breast cancer cohorts (Casadei S et al. Cancer Res. 2011 Mar;71:2222-9; Antoniou AC et al. N. Engl. J. Med. 2014 Aug;371:497-506; Decker B et al. J. Med. Genet. 2017 Aug). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
