The lysine at codon 745 is replaced by arginine, an amino acid with highly similar properties. This alteration has been identified in at least two patients with cancer (Aries IM et al. J. Exp. Med. 2018;215(12):3094-3114, DeRycke MS et al. Mol Genet Genomic Med. 2017;5(5):553-569). This alteration was found to be functionally normal in a homology-directed DNA repair (HDR) assay (Wiltshire T et al. Genet. Med., 2020 03;22:622-632). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
