The p.T413K variant (also known as c.1238C>A), located in coding exon 4 of the PALB2 gene, results from a C to A substitution at nucleotide position 1238. The threonine at codon 413 is replaced by lysine, an amino acid with similar properties. This alteration was found to be functional in a homology-directed DNA repair (HDR) assay (Wiltshire T et al. Genet. Med., 2020 Mar;22:622-632). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
