This variant is present in population databases (rs776110440, gnomAD 0.0009%). This missense change has been observed in individual(s) with breast cancer (PMID: 27616075). ClinVar contains an entry for this variant (Variation ID: 188076). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.S29G variant (also known as c.85A>G), located in coding exon 2 of the PALB2 gene, results from an A to G substitution at nucleotide position 85. The serine at codon 29 is replaced by glycine, an amino acid with similar properties. In a study of 581 German high-risk BR and/or OV cancer patients, this variant was detected in one family (Kraus C et al. Int. J. Cancer, 2017 Jan;140:95-102). This alteration was also detected in 1/5589 German BRCA1/2-negative probands diagnosed with breast cancer (Hauke J et al. Cancer Med, 2018 Apr;7:1349-1358). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
