This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 89 of the ZNF423 protein (p.Arg89His). This variant is present in population databases (rs745597535, gnomAD 0.05%). This missense change has been observed in individual(s) with clinical features of Joubert syndrome (PMID: 26539891). ClinVar contains an entry for this variant (Variation ID: 402222). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ZNF423 protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change does not substantially affect ZNF423 function (PMID: 32925911).
