The p.K440E variant (also known as c.1318A>G), located in coding exon 9 of the CDH1 gene, results from an A to G substitution at nucleotide position 1318. The lysine at codon 440 is replaced by glutamic acid, an amino acid with similar properties. This variant was detected in multiple individuals with no reported features of CDH1-related diffuse gastric and lobular breast cancer (DGLBC) (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
