Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.9e-05 in 1614058 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in CDH1 causing hereditary diffuse gastric cancer (1.9e-05 vs 2.8e-05), allowing no conclusion about variant significance. c.1370C>T has been reported in the literature without strong evidence for or against pathogenicity (example: Bodian_2014, Garcia-Pelaez_2022). These report(s) do not provide unequivocal conclusions about association of the variant with hereditary diffuse gastric cancer.
