This missense variant replaces glutamic acid with aspartic acid at codon 482 of the CDH1 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with CDH1-related disorders in the literature. This variant has been identified in 1/31388 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.E482D variant (also known as c.1446A>T), located in coding exon 10 of the CDH1 gene, results from an A to T substitution at nucleotide position 1446. The glutamic acid at codon 482 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This variant has not, to our knowledge, been published in the literature as pathogenic or benign. CDH1 Glu482Asp was not observed in large population cohorts (NHLBI Exome Sequencing Project, The 1000 Genomes Consortium 2015, Lek 2016). Since Glutamic Acid and Aspartic Acid share similar properties, this is considered a conservative amino acid substitution. CDH1 Glu482Asp occurs at a position that is conserved across species and is located in the Cadherin 3 domain (Brooks-Wilson 2004, Figueiredo 2013).
