In a Japanese case-control study, this variant was not reported in any breast cancer cases, but was observed in unaffected male controls (Momozawa 2018). CDH1 Asp644Asn was not observed at a significant allele frequency in large population cohorts (Lek 2016). This variant is located in cadherin 5 of the extracellurlar domain (Brooks-Wilson 2004, Figueiredo 2013, UniProt). In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function.
