Variant summary
Our verdict is Uncertain significance. Variant got 4 ACMG points: 4P and 0B. PM2PP3_Moderate
The NM_001361.5(DHODH):c.95G>T(p.Gly32Val) variant causes a missense change involving the alteration of a conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★).

The missense variant p.G32V in DHODH (NM_001361.5) has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.G32V variant is novel (not in any individuals) in gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. There is a moderate physicochemical difference between glycine and valine. The p.G32V missense variant is predicted to be damaging by both SIFT and PolyPhen2. The glycine residue at codon 32 of DHODH is conserved in all mammalian species. The nucleotide c.95 in DHODH is predicted conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance. -
