This alteration results from a G to A substitution at nucleotide position 389, causing the arginine (R) at amino acid position 130 to be replaced by a lysine (K). This sequence change replaces arginine, which is basic and polar, with lysine, which is basic and polar, at codon 130 of the CCDC78 protein (p.Arg130Lys). This variant is present in population databases (rs375016420, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with CCDC78-related conditions. ClinVar contains an entry for this variant (Variation ID: 583302). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt CCDC78 protein function with a negative predictive value of 80%.
