Variant summary
Our verdict is Uncertain significance. The variant received 0 ACMG points: 2P and 2B. PM2BP4_Moderate
The NM_018975.4(TERF2IP):c.16G>A(p.Asp6Asn) variant causes a missense change. The variant allele was found at a frequency of 0.00000344 in 1,454,696 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. D6Y) has been classified as Uncertain significance.

The p.D6N variant (also known as c.16G>A), located in coding exon 1 of the TERF2IP gene, results from a G to A substitution at nucleotide position 16. The aspartic acid at codon 6 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
