IP3 and DAG are second messenger molecules important for transmitting signals from growth factor receptors and immune system receptors across the cell membrane. This variant is listed in the Genome Aggregation Database (gnomAD) with a frequency of 0.06 percent in the European Non-Finnish population (identified on 71 out of 126,704 chromosomes). The proline at position 236 is highly conserved up to baker’s yeast considering 13 species (Alamut v2.10) and computational analyses of the effects of the p.Pro236Leu variant on protein structure and function provide conflicting results (SIFT: tolerated, MutationTaster: disease causing, PolyPhen-2: probably damaging). Altogether, there is not enough evidence to classify the p.Pro236Leu variant with certainty. - |
