Variant summary: PMM2 c.385G>C (p.Val129Leu) results in a conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 199802 control chromosomes. c.385G>C has been reported in the literature in compound heterozygous individuals affected with Congenital Disorder Of Glycosylation Type 1a (Wurm_2007). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.385G>A, p.Val129Met), supporting the critical relevance of codon 129 to PMM2 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 16941129). ClinVar contains an entry for this variant (Variation ID: 1068915). Based on the evidence outlined above, the variant was classified as likely pathogenic. -
