The p.R350L variant (also known as c.1049G>T), located in coding exon 12 of the FANCA gene, results from a G to T substitution at nucleotide position 1049. The arginine at codon 350 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.

This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 350 of the FANCA protein (p.Arg350Leu). ClinVar contains an entry for this variant (Variation ID: 439689).
