This gene is found in a cluster of myosin heavy chain genes on chromosome 17. A mutation in this gene results in inclusion body myopathy-3. This alteration results from a C to T substitution at nucleotide position 3709, causing the leucine (L) at amino acid position 1237 to be replaced by a phenylalanine (F). The L1237F variant in the MYH2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The L1237F variant is observed in 6/66694 (0.009%) alleles from individuals of non-Finnish European background, in the ExAC dataset (Lek et al., 2016). The L1237F variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret L1237F as a variant of uncertain significance. -
