Studies suggest two alternately used promoters drive tissue-specific expression. This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 67 of the PMP22 protein (p.Ala67Thr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of PMP22-related conditions (PMID: 12796555, 29108667). ClinVar contains an entry for this variant (Variation ID: 8443). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect PMP22 function (PMID: 26102530, 33933451). This variant disrupts the p.Ala67 amino acid residue in PMP22. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 10330345, 11920834; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
