This sequence change replaces histidine, which is basic and polar, with arginine, which is basic and polar, at codon 2309 of the PRPF8 protein (p.His2309Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with retinitis pigmentosa (PMID: 18695108). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 3355). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt PRPF8 protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects PRPF8 function (PMID: 28515276). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
