This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with an NF1-related disease. In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces valine with phenylalanine at codon 54 of the NF1 protein (p.Val54Phe). The valine residue is highly conserved and there is a small physicochemical difference between valine and phenylalanine. -

The p.V54F variant (also known as c.160G>T), located in coding exon 2 of the NF1 gene, results from a G to T substitution at nucleotide position 160. The valine at codon 54 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
