The p.I117S variant (also known as c.350T>G), located in coding exon 4 of the NF1 gene, results from a T to G substitution at nucleotide position 350. The isoleucine at codon 117 is replaced by serine, an amino acid with dissimilar properties. This alteration was identified in an individual with a clinical diagnosis of neurofibromatosis type 1 (NF1) (Ars E et al. Hum Mol Genet, 2000 Jan;9:237-47). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The variant is moderately destabilizing to the local structure (Ambry internal data). Based on the available evidence, the clinical significance of this alteration remains unclear.
