Mutations in this gene have been linked to neurofibromatosis type 1, juvenile myelomonocytic leukemia and Watson syndrome. The W223X variant in the NF1 gene has been reported in at least one individual with clinical features of neurofibromatosis type 1 (Pasmant 2015). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The W223X variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Based on currently available evidence, we consider W223X to be a pathogenic variant. -

The p.W223* pathogenic mutation (also known as c.669G>A), located in coding exon 7 of the NF1 gene, results from a G to A substitution at nucleotide position 669. This changes the amino acid from a tryptophan to a stop codon within coding exon 7. This alteration was reported inan individual with neurofibromatosis (PasmantE et al.Eur. J. Hum. Genet. 2015 May;23(5):596-601). In addition to the information presented in the literature, since premature stop codons are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294).<br /> . -
