The leucine at codon 380 is replaced by proline, an amino acid with similar properties. This variant has been detected in multiple individuals who met diagnostic criteria for neurofibromatosis 1 (NF1), including three individuals from the same family (Cunha KS et al. Genes (Basel), 2016 Dec;7:) and one unrelated individual with early-onset breast cancer before the age of 50 (Frayling IM et al. J Med Genet, 2019 04;56:209-219). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic. - |
