It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with neurofibromatosis type 1 (PMID: 15060124, 18546366, 29618358, 31717729). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 404540). Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 18546366). This mutation has been identified in an individual with clinically diagnosed neurofibromatosis type 1 (NF1) with an optic pathway glioma (Hutter S et al. Hum. Genet., 2016 May;135:469-75). This mutation has also been identified in another individual with clinically diagnosed or suspected NF1 and was found to cause aberrant splicing leading to the skipping of exon 10b (Pros E et al. Hum. Mutat., 2008 Sep;29:E173-93). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as a disease-causing mutation. - |
