Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has been observed in individual(s) with breast cancer (PMID: 30287823). This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with leucine at codon 652 of the NF1 protein (p.Arg652Leu). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and leucine.

The p.R652L variant (also known as c.1955G>T), located in coding exon 17 of the NF1 gene, results from a G to T substitution at nucleotide position 1955. The arginine at codon 652 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
