The p.L691R variant (also known as c.2072T>G), located in coding exon 18 of the NF1 gene, results from a T to G substitution at nucleotide position 2072. The leucine at codon 691 is replaced by arginine, an amino acid with dissimilar properties. This variant was reported in individual(s) with features consistent with neurofibromatosis type 1 (NF1) (Balla B et al. J. Mol. Neurosci., 2014 Jun;53:204-10). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
