This variant has been observed to be de novo in an individual affected with neurofibromatosis type I (PMID: 27322474). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Nucleotide substitutions within the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Experimental studies have shown that this variant causes abnormal splicing which creates a transcript that is lacking of exon 19 and results in a frameshift protein (PMID: 18546366, 27322474). Exon 19 is also known as exon 14 in the literature. For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamic acid with aspartic acid at codon 775 of the NF1 protein (p.Glu775Asp). The glutamic acid residue is moderately conserved and there is a small physicochemical difference between glutamic acid and aspartic acid. This variant also falls at the last nucleotide of exon 19 of the NF1 coding sequence, which is part of the consensus splice site for this exon. -
