The tryptophan at codon 777 is replaced by arginine, an amino acid with dissimilar properties. This mutation has been identified in multiple individuals with a clinical diagnosis of neurofibromatosis type 1 (NF1) (Cai Y et al. J. Dermatol. Sci., 2005 Aug;39:125-7; Evans DG et al.
