The NF1 c.2410-12T>C variant is predicted to interfere with splicing. This variant was reported as a variant of uncertain significance in an individual with a suspected RASopathy diagnosis (Table S2 - Witkowski et al. 2020. PubMed ID: 32107864). This variant has not been reported in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. In ClinVar this variant has conflicting interpretations of pathogenicity of likely benign and uncertain (https://ncbi.nlm.nih.gov/clinvar/variation/229062/). However, the use of computer prediction programs is not equivalent to functional evidence, and therefore the clinical significance of this variant is uncertain. -
