The p.R997S variant (also known as c.2991G>T) is located in coding exon 23 of the NF1 gene. The arginine at codon 997 is replaced by serine, an amino acid with dissimilar properties. This change occurs in the first base pair of coding exon 23. This variant was determined to be de novo in at least one individual with features consistent with Neurofibromatosis type 1 (Ambry internal data; personal communication). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic. -
