The arginine at codon 1000 is replaced by proline, an amino acid with dissimilar properties. This variant was reported in multiple individuals who met clinical criteria for Neurofibromatosis type 1 (NF1) (external communication). Based on internal structural analysis, this variant is highly destabilizing to the local structure (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the supporting evidence, this alteration is interpreted as a disease-causing mutation. - |
